This book chapter reviews recent all-atom simulations that aim at characterizing the monomeric structure of intrinsically disordered peptides implicated in protein aggregating diseases. Simulations reveal that peptides that appear to be disordered based on experimental studies that make use of ensemble-averaging techniques, in fact possess local secondary structure that may play a role in initiating aggregation.
